摘要:
采用单因素分组法,以脱蛋白率和多糖保留率为评价指标,研究并比较壳聚糖和ZTC1+1Ⅱ型絮凝剂对猴头菇总多糖提取液的纯化工艺。通过单因素试验,系统考察絮凝剂用量、絮凝温度、作用时间对纯化工艺的影响,确定了两种絮凝剂纯化的最佳工艺参数,最后还对比了两种絮凝剂纯化的猴头菇总多糖溶液对1,1-二苯基-2-三硝基苯肼自由基(DPPH·)的清除能力。结果表明:壳聚糖絮凝效果优于ZTC1+1Ⅱ型絮凝剂,当絮凝温度为50 ℃、壳聚糖用量为0.8 mL/g、絮凝时间为2.0 h时,脱蛋白率为38.52%,多糖保留率为94.65%,并且壳聚糖絮凝纯化后的提取液清除DPPH·的能力优于ZTC1+1Ⅱ型絮凝剂。
Abstract:
The purification of the polysaccharide solution obtained from Hericiumerinaceus by chitosan and ZTC1+1Ⅱ flocculants were studied with the deprotobeinizing rate and the polysaccharides retention rate as the evaluation indicators.The effects of factors such as dosage of flocculant,flocculation time,and flocculation temperature on the flocculation were investigated by single-factor experiments,and the optimum parameters were obtained.Results showed that the flocculation effect of chitosan was better than ZTC1+1Ⅱ flocculants,and the optimum flocculation parameters were as follows:the temperature of 50 ℃,the amount of chitosan of 0.8 mL/g(flocculant volume/hericium amount)and flocculation time of 2.0 h.Under the optimum conditions,the deproteinizing rate was 38.52% and the polysaccharides retention rate was 94.65%.1,1-diphenyl-2-picrylhydrazyl-radical(DPPH·)-scavenging assay was also carried out to evaluate the bioactivity of polysaccharide solution of hericium purified by chitosan and ZTC1+1Ⅱ flocculant with a better bioactivity by chitosan.
